If your going to that much trouble to swap a tranny, i'd opt for a ZR6. 6-speed would be amazing with lots of power. T56 would be even better, but im not sure how it'd be done on a 4x4 unit. I've only seen T56's on a 2x4.

Or thats the plan. Might put the big block in the blazer and small block in the truck.

If I go with the 5R110, I'll use a PCS trans controller and use a PCM strategy from a manual. So the PCM doesnt control the transmission. Transfercase will be the easy part. Just a matter of swapping the main shaft to match the SD. I've already done it one on a Bw1346. I had to do it on my SCAB when I put the billet input/output shafts in my E4OD.
