A/C Clunk
95 f150 300 i6 4wd a/c
When I turn on my air conditioning on (after a few days of not using it) it makes a loud single "clunk" when I turn it on.
Works great otherwise, and doesn't do this if I use it regularly from day to day.
Anything of concern here?

likely clutch on the A/C compressor. It should "unclunk" when the A/C is turned off.
It will clunk on and off throughout it's life until one day it will not and smoke/fire will fill the ring of the A/C compressor and eventually become a full fledge fire. Like mine did. 20 plus years of calm clunks.
Cost me $1200 to tow it 600 miles to my new home, while we rode with the moving van. Great weekend. Not so much.

Next time it sits long enough, open the hood before starting the engine and have an assistant crank it & turn the A/C on while you watch.
But my guess is that it's a door in the HVAC ducts moving to its selected position, after the vacuum has bled down over the previous few days. You can check the vacuum lines & reservoir for leaks, but if it takes days, you'll probably never find it. Read these pages & follow the relevant links on them:
