If MPG"s are your concern, it doesn't matter which one you consider.....it's not even noticeable. I can tell you I average 17.5 mpg in the Chicago burbs driving back and forth to work on all city streets in the summer. Highway MPG's suck though for my driving, but it's a 3.73 gear and I always go 75mph. I get ~17mpg highway because of this in the summer.
To get the 21mpg rating I have to do 60mph and I'm simply incapable of doing that on a long road trip. Mine is an Ecoboost in my avatar, and I loved the twin turbo idea in a pickup. I'm also mechanically inclined.
Each one has pro's and con's. The 5.0L is a great motor too, and obviously much simpler and less costly to maintain in the long run more then likely.
TOWING......there's no competition, the Ecoboost destroys all competitors trucks. Power Potential with a simple tune , another area it will excel against all competition. But MPG"s.......negligible.

^^x2. Look, we all know the Eco will tow better than a 5.0, but the 5.0 is no slouch. I have a 5.0 and love it. I have never wanted or needed more power. I will say that I am interested in the ecoboost and think I will consider it in my next truck. Guys are getting crazy numbers with just a tune.

In town every truck is nearly identical and so is every car unless it's a hybrid. So I would base my decision on that. They are both generally reliable. The Eco has more power everywhere and will kill the 5.0 is low end torque. But even used the Eco commands a premium so you need to decide if you want the more powerful engine with the same city mog and a couple better mpg on hwy or get the cheaper less powerful engine. No different than choosing between an accord 4 cylinder and the v6.
Both engines are far from what I consider fast. I bought a pickup, not a sports car.

Eco will out tow the 5.0, no competition. Not saying the 5.0 won't tow the same load though. Me living at a higher altitude and towing 6k+, it wasn't even close. Eco tows here way better than the 5.0.

I wanted the ecoboost, but i end up with with a 5.0, because of the what I wanted on the truck. I think either engine is just as good as the other. As far as gas mile goes, it all depends on how heavy your foot is.
