It really depends..... I have a 5500lb boat/trailer that I launched& retrieved 15 times last year in 2wd, even with seaweed on the ramp it was ok. I also have a 14 foot fishing boat that weighs less than 1000lbs on trailer, and needed 4wd to get out because of the ramp condition.
2wd should be fine, but you need to carefully study the ramp first. If its tidal and you get the rear wheels on slime, forget it... But if you use your head and ensure you have traction it doesn't matter if your boat weighs 200, 2000 or 10000 lbs 2wd will be fine.
The 2011+ trucks have a very nice wheel slip control, even with an open diff. I have limited slip in addition to that.

It does just depend. My dad has a 1974 (has very thick fiberglass) 27' boat with a 460 in the back and he made it up the ramp fine but when I pulled my tiny jet ski on a trailer up the same ramp I was spinning tires the whole way. But I also had near bald tires and wasn't trying to keep my traction.

I have pulled big and small boats for year and have never even had 4wd and never been stuck on the ramp. Of course I have a few ramps I won't go to because of the ramp conditions, but all in all it is just being smart and using the throttle correctly.
