Even just doing the tuner alone will give very noticeable results. I have a Roush exhaust, but I don't think it did anything other than make the truck sound nice.
The track I am at is notoriously slow for a variety of reasons... so my numbers sound slow compared to the Atco types. Stock tune with 26in rims I went 15.26 @ 94mph. With stock 17 inch wheels, and 5 Star 91 octane tune I ran 14.41 @ 98mph. For some comparison, there was a new RCSB Ram Hemi (390hp) that had headers, full exhaust, gears, cai, and a tune... his best time of the night was 14.88.

Atco is a track in New Jersey with only 90ft elevation. There's always a lot of fast times coming from that track. The track I go to (Saskatchewan International Raceway) is 1500ft elevation, and when I made my passes the density altitude was 3500ft. Cooling the truck down seems to make a significant difference. I made 5 passes in a row that were bracket racer consistent between 14.55-14.61, then let the truck cool off for an hour, then ran the 14.41.

I need some advice, this f-150 is the first real truck I've ever owned. I'm looking to start installing performance mods, where do I start to get the best throttle response and fuel economy? I hear slot of talk about the boss intake manifolds, should I start there?!
