Got a little thump when leaving a stop
2010 XL regular cab, 63,000 mi., full size bed, 2X, 4.6 auto....I have a slight thump most of the time that I pull away from a stop. It feels like something has to catch up to the drivetrain. No noise and the truck has no rattles. I thought it might be a loose spare tire, nope! Could it be the rear brake pads? And no, their is nothing in the bed sliding around. Bought it used about two months ago. It is not getting any worse and of coarse the used car dealership said it was normal. Due to knee surgery I haven't been under it yet.

Mine does the same thing. I feel it may have something to do with the "Hill Start Assist".
After I back my boat in the water and I put the truck in gear to pull away, there is a brief second where it feels like the brake is still engaged, then releases with a slight noise. My truck seems to do the same thing each time I stop, regardless of whether I'm on a hill or not, just to a lesser degree.
I think its normal and am not concerned about it.

There is a dealer fix for this issue. I had mine done a few months ago. They replaced a seal on the spline. As long as I was using the truck for towing, it would "fix" itself but after a couple weeks of no towing, the thump would return.
