Cost to replace trans pan gasket and trans electrical harness seal
My 2010 5.4L has leaked 3 quarts of trans fluid since Friday. I took it to a trans shop this morning and they diagnosed it as both the pan gasket and the seal for the electrical harness connector. They quoted about $140 in parts and $400 in labor to drain it, replace the filter, gasket, and seal, and refill it. Am I being hosed or is it really this much work?

That sounds too high....If it is the same connector and seal that started leaking in my 2010, they have to drop the pan and replace that seal from the inside of the transmission. I can't remember, but i think i paid about $200-250 total at the Ford dealership.
Mine was barely leaking and not even hitting the ground yet, only got the undercarriage a little moist. I replaced it anyway, because I didn't want to risk it failing on the highway and dumping all my fluid at once.

You can do it all yourself for about $120.

This is so odd.
I got 4 quotes today:
Chain trans shop: $500
Independent auto shop: $800
Independent auto shop: $500
Ford dealership: $850
They all quote 4 to 4.5 hours and scoffed at the idea of charging $250 for this repair.
This for a pan gasket, bulkhead seal, filter, and full flush on a 2010 Ford F-150 with 5.4L V8 and 6-speed trans.
I got 4 quotes today:

Wow, those are high. Had mine done at the Ford dealership a few years ago for around $300. If I didn't have an extended warranty that paid for most of it, I would have done it myself.

Mine was leaking aswell, my quotes were over $450. Fordtechmakuloco on YouTube has some great videos on it. I do lots of my own maintenance, but had never done anything with a transmission before, yet I tackled it myself.
It took me about 2 hours including clean up and refilling, and around 150-200 Canadian for the oil, filter, bulkhead connector and a small bottle pump.
Couple things I noticed in your quote that I believe arent necessary:
Pan gasket- it metal and reusable, took 2 mins with a rag to clean it (replace it its damaged) mine had 140k on it and was fine
Flush - apparently there's a thermostatic valve somewhere in the transmission, that has to be brought up to temperature in order to flush and recirculate the fluid.
if you're willing to tackle the job it's really as simple as:
disconnect battery
clean the area
Loosen bolts in pan, leave a couple in, drain the pan
pull the retaining clip out of bulkhead connector, push a new one in, make sure clip is back in fully
Insert a new transmission filter ( just friction fits in a tube)
reinstall pan gasket + pan
refill fluid high as possible cold, let it idle and warm up, fill to the hashmarks
Like I said, check out the YouTube videos, see if you want to do it yourself to save a few hundred dollars
