Default Choices, choices...

I have the 300 in my truck and have Hooker headers on it. I plan on putting a 4 barrel carbon intake on it. My question is, will a 2 or 4 barrel be better?

It depends what you want. A spreadbore 4 barrel will give you the best performance because the smaller primaries will allow better fuel atomization because of the increased airflow through the venturies, and also better throttle response. That said many of the guys with sixes and old 289 like the 390 CFM square bore and its an ideal setup for the 300. The best part of the 4 v is the kicking in of the secondaries. If you like playing with carbs go for four.
If your looking for trouble free setup and simplicity the popular 2 barrels will fill the bill. WOT performance won't be much different if the CFM is equal and they are generally cheaper.

You'll get worse mileage with a 2 barrel.
