Can leaking fluid cause a loss of first gear?

Leaking itself won't but low fluid will cause irratic shifting. Fix the leak, fill with fluid and see if it's still present.

Either end or pan/gasket. Go under and look. If you can't find it, clean everything up then check periodically.

Anything that doesn't seem normal. Missing gears, delayed..etc. Start with the easy stuff and work your way up. How low was it? Noticeable puddles under the truck?

With converter and all 400. Just had it rebuilt about 4 months ago. Works great.
