setting input sensitivity

Default setting input sensitivity

Ok, I've got a pioneer avh-p4400bh (4 volt pre-out) with the front outputs going into the front channels of a jl audio slash 300/3, the rear outputs going into the rear channels of the same amp and the sub outputs going into a jl audio 250/1. I have a test cd with the appropriate sine waves and a digital voltage meter. My question is, when I disconnect the speakers and connect the meter do I have to do each speaker independently to properly set the input sensitivity or can I just do 1 speaker and if so, which 1?

I would do the front pair, rear pair and sub separately.
