Question P030x misfire codes

Concerning the P0301 thru P0308 cylinder misfire OBD codes. What exactly turns these codes on. Possible causes in my mind are:
Low compression
Lack of combustion
No spark
What information I am seeking here is "What sensor or combination of sensors data does the computer digest to come up with a cylinder misfire. I just rebuilt a 1997 Ford Escort and after 50 miles got a P0304 code (#4 cylinder misfire). I hooked up the #4 plug wire and no more P0304. So.....How did the computer sense the misfire? The only sensors I am aware on the Escort head are the crank sensor (spark) and the cam sensor (not real sure what it does)? No big deal, JUST CURIOUS?
